چکیده

Although so much stress is laid on the physiological reactions of bacteria in their identification and classification, little investigation (except to distinguish aerobic from anaerobic forms) has been devoted to the fundamental process of respiration in these organisms. In order to throw some light on the nature of this process and to compare the results with those obtained with other plants and with animals, a series of studies upon the respiration of bacteria has been undertaken. The present paper describes the results of some experiments on the effects of ether. The organism selected for experimentation was Bacillus subtilis, isolated from water and inoculated upon agar-agar daily during the month previous to experimentation, so that a pedigreed culture was at hand. Several other organisms were tried; namely, Bacillus mycoicles, Staphylococcus pyogenes albus, Staphylococcus citreus, Bacillus typhosus; but Bacillus subtilis was preferred because it was easier to handle. Bacteria were incubated for 16 to 18 hours at 37°C. and were then washed into a test-tube with a few cc. of tap water containing indicator. A thick emulsion was made by repeatedly drawing the liquid into, and ejecting it from a pipette which was drawn out to a very fine point. The respiration of the bacteria was tested in boiled tap water. The boiling rendered the water alkaline by driving off the COs. To each 2 cc. of solution was added one drop of a 0.01 per cent aqueous solution of phenolsulfonephthalein. The color of the solution was compared with the colors of a set of standard buffer solutions, made according to SSrensen's directions with KH~PO4 and Na2HPO4, hay193

COMPARATIVE STUDIES ON RESPIRATION. X. Toxic AND ANTAGONISTIC EFFECTS OF MAGNESIUM IN RELATION TO THE RESPIRATION OF BACILLUS SUBTILIS

The behavior of magnesium is of special interest since in studies on antagonism magnesium has been found to occupy an intermediate position between the monovalent and the bivalent metals. In many cases it can act as an antagonist to calcium as well as to sodium. As no investigations on respiration have been undertaken from this point of view, the writer has made some experiments on this subject...

Evaluation of the effects of diethyl ether and methanol extracts of carpet anemone (Sarcophyton spp.) and starfish (Pentaceraster spp.) on E. coli and Bacillus subtilis bacteria

Marine environment is the main source of natural bioactive products whose chemical-structural properties are not similar with other natural products of plants and other inland organisms. The present study was carried out to evaluate antibiotic properties of diethyl ether and methanol extracts of two marine species including starfish and carpet anemone. The samples of starfish and carpet anemone...

Comparative Studies on Respiration

1. In relatively low concentrations of NaCl, KCl, and CaCl(2) the rate of respiration of Bacillus subtilis remains fairly constant for a period of several hours, while in the higher concentrations, there is a gradual decrease in the rate. 2. NaCl and KCl increase the rate of respiration of Bacillus subtilis somewhat at concentrations of 0.15 M and 0.2 M respectively; in sufficiently high concen...

Cisplatin cytotoxicity is dependent on mitochondrial respiration in Saccharomyces cerevisiae

Objective(s): To understand the role of mitochondrial respiration in cisplatin sensitivity, we have employed wild-type and mitochondrial DNA depleted Rho0 yeast cells. Materials and Methods: Wild type and Rho0 yeast cultured in fermentable and non-fermentable sugar containing media, were studied for their sensitivity against cisplatin by monitoring growth curves, oxygen consumption, pH changes ...

The effect of 6-deoxyclitoriacetal from Clitoria macrophylla Wall. on rat liver mitochondrial respiration and ATPase activity

A Thai plant Clitoria macrophylla Wall. was claimed to possess some pharmacological activities. The phytochemical studies reported a rotenoid compound, 6-deoxyclitoriacetal, which exhibits the cytotoxic effect in several cell line experiments. This report was to study the effect of 6-deoxyclitoriacetal on rat liver mitochondria, which may associated to its cytotoxic phenomena. Mitochondrial sus...
